【Objective】

This study aimed to analyze and evaluate the effects of sweet corn slurry with different solid content on the physical stability and sensory quality of a total nutrition emulsion of food for special medical purposes (FSMP), to determine the appropriate concentration of sweet corn slurry for such a product, and to develop a sweet corn slurry-based emulsion-type FSMP product, thus providing a reference for the research and development of FSMP products formulated from agricultural products and natural ingredients.

【Method】

Sweet corn slurries with solid content of 3%, 4%, 5%, 6%, and 7% were respectively used as the aqueous phase matrix, and compounded with maltodextrin, sodium caseinate, soybean oil, vitamins, and minerals, followed by homogenization and sterilization to obtain a total nutrition FSMP emulsion. The stability of this emulsion-type FSMP was assessed in terms of such evaluation indexes as centrifugal sedimentation rate, particle size distribution, microscopic morphology, turbiscan stability index (TSI), steady-shear rheological properties, sensory scores and electronic tongue information, combined with short-term storage tests.

【Result】

With the increase in solid content in sweet corn slurries, the centrifugal sedimentation rate of the emulsion gradually increased, which accompanied by a higher proportion of large particles and the appearance of multiple peaks in the particle size distribution. Microscopic observations revealed an increasing number of large particle fragments, with slight flocculation in the emulsions prepared with slurries with 6% and 7% sweet corn solid content. The TSI values decreased initially and then increased, reaching a minimum at a sweet corn solid content of 4%. Apparent viscosity increased with the solid content, showing a shear-thinning pseudoplastic flow behavior; the emulsions prepared with slurries with 3% and 4% sweet corn solid content had relatively low viscosities and good flow properties. Sensory evaluation and electronic tongue analysis showed that the emulsion prepared with the slurry with 4% sweet corn solid content had the highest score and the best taste, and no creaming or sedimentation was observed after 21 days of short-term storage.

【Conclusion】

Considering all evaluation indices, the emulsion prepared with the slurry with a 4% sweet corn solid content achieved a uniform and stable system with favorable flow properties and a pronounced sweet corn flavor, making it suitable as the aqueous phase matric for an emulsion-type FSMP. The use of this emulsion could not only endow FSMP products with a natural flavor but also improve the nutrition and energy levels of the products.

The changes of volatile flavor compounds in three varieties of sweet corn, “Zhongtian 9”, “Zhuyutian 8” and “Zhuyutian 1”, were investigated during the ripening process. The volatile flavor compounds were identified by headspace solid phase microextraction and gas chromatography-mass spectrometry (HS-SPME-GC-MS) through NIST14 mass spectral database searching and retention index analysis as well as by gas chromatography-olfactory (GC-O), and were quantified by the internal standard method, in order to construct a volatile flavor profile and evaluate the aroma characteristics of sweet corn. The results showed that 61 volatile flavor compounds were detected across the three varieties during the ripening process. Maturity had significant effects on the type and content of volatile flavor compounds in sweet corn. The contents of ethyl nonylate, 1-heptanol and 1-hexanol significantly decreased with increasing maturity, while the contents of styrene, toluene, ethylbenzene and m-xylene significantly increased. Arenes were mainly accumulated in sweet corn during the maturation process. In terms of odor characteristics, the intensity of fatty aroma increased obviously with maturity, while the intensity of grassy aroma decreased obviously. There were significant differences among varieties in the composition and content of volatile flavor compounds. The number and amount of volatile flavor compounds in “Zhongtian 9” were significantly greater than those in the other two varieties at maturity, and the overall flavor intensity was the highest.

The effects of blanching followed by spray drying on volatile flavor compounds in sweet corn were investigated. The volatile flavor compounds in fresh, blanched and spray-dried sweet corn were analyzed by headspace solid phase microextraction coupled with gas chromatography-mass spectrometry (HS-SPME-GC-MS). The qualitative analysis was carried out by comparison of the spectra for the chromatogram peaks with those in the NIST14 database library and comparison of retention index (RI), and the quantitative analysis by an internal standard method. Gas chromatography-olfactometry (GC-O) was used to analyze the composition of odor-active compounds and overall flavor profiles. The results showed that a total of 58 volatile flavor compounds were detected in fresh, blanched and spray-dried samples. Blanching and spray drying had significant effects on the types and contents of volatile flavor compounds in sweet corn. After blanching treatment, the contents of ketones, aromatic compounds, terpenes and aldehydes were decreased, and the total content of volatile flavor compounds decreased by 36.60%. After spray drying, the total content of volatile flavor compounds was increased by 19.21 and 30.30 times compared with those of fresh and blanched sweet corn, respectively. Among them, the contents of aldehydes, ketones, alcohols and heterocyclic compounds were increased, and new substances such as dimethyl thioether and 2-acetyl-1-pyrroline were detected. Principal component analysis (PCA) showed that the decreases in the contents of 2-nonenal (E), 3-ethyl-2-methyl-1,3-hexadiene, octanal and D-limonene were closely related to blanching, while the increases in the contents of dimethyl thioether, 2-acetyl-1-pyrroline, 2-methylpropanal, 3-methylbutanal, 2-methylbutanal 2-methyl-(E)-2-butenal compounds were highly correlated with spray drying. Thirty-three odor-active compounds were detected by GC-O, 10 of which had odor intensity value (OIV) equal to or larger than 3. Flavor profile analysis showed that the intensity of fruity aroma was the highest in fresh sweet corn, followed by floral, nutty and green aroma. After blanching treatment, the overall aroma intensity was decreased and in particular, the grassy aroma intensity was decreased significantly. After spray drying, the overall aroma intensity was significantly increased, the intensity of nutty and green aroma was significantly enhanced, and a strong barbecue aroma appeared.

【Objective】

Tea saponin is a naturally active substance with wide application prospective, and its purity is the key factor limiting its application and value. In this study, the optimal conditions for ultrasonic-assisted two-phase extraction (UAATPE) of tea saponin from camellia meal were explored, and the possible extraction mechanism was discussed in order to develop an efficient, high-purity extraction technology, so as to provide a technical guidance for high-value utilization of camellia meal.

【Method】

On the basis of the single factor experimental results, the key factors affecting the yield of tea saponin were screened by Plackett Burman design, and the extraction process was optimized by Box-Behnken design. The extraction efficiency of tea saponin using the UAATPE method was evaluated by comparing the extraction yield and purity to the traditional ethanol extraction method and water extraction method. Scanning electron microscopy (SEM) was employed to analyze the microstructure of camellia meal, and the possible mechanism of extraction by the UAATPE method was discussed.

【Result】

The key factors affecting the yield of tea saponin were mass fraction of ethanol, mass fraction of ammonium sulfate, and ultrasonic time. The process parameters optimized by Box-Behnken design were as follows: an ethanol content of 27.50% (W/W), an ammonium sulfate content of 19.60% (W/W), a liquid-solid ratio of 50∶1, an ultrasonic time of 32 min, an ultrasonic power of 300 W and an extraction temperature of 60℃, and the yield of tea saponin at (26.21±0.54)% was achieved under the optimized conditions. Compared with the traditional method of ethanol extraction, the purity of tea saponin extracted by the UAATPE method increased by 6.57% (P<0.05), although the difference of the yield of tea saponin extracted by these two methods was not significant. Compared with the traditional method of water extraction, the yield and purity of tea saponin extracted using the UAATPE method were increased by 17.74% and 40.23%, respectively (P<0.01). The microstructure showed that owing to the effect of the camellia meal treated by the UAATPE method, the ultrasonic cavitation accelerated the tissue damage of the camellia meal, producing a large number of voids and strong surface shrinkage, which effectively promoted the release of tea saponin in the camellia meal. In the process of the UAATPE extraction, the tea saponin first entered the bottom phase with high electrical conductivity through solid-liquid extraction in the two-phase water system, and then moved to the top phase with high polarity through liquid-liquid extraction, achieving primary purification and improving the purity of tea saponin.

【Conclusion】

The UAATPE method could significantly improve the yield and purity of tea saponin, thus providing a new novel method for the efficient utilization of camellia meal.
