Construction of A Comprehensive Evaluation System and Screening of Cold Tolerance Indicators for Cold Tolerance of Cotton at Seedling Emergence Stage

【Objective】

In this study, the purpose was to comprehensively evaluate the cold tolerance of cotton varieties (lines) at seedling emergence stage, establish a reliable evaluation model, screen and identify indicators, and provide a simple and effective evaluation method for the selection and identification of cold-tolerant varieties in cotton.

【Method】

200 upland cotton varieties (lines) were used to test hypocotyl length, root length and 100-grain weight, etc. under three treatments of constant chilling (CC), diurnal variation of chilling (DVC) and normal conditions. A combination of integrated cold tolerance coefficient difference analysis, frequency analysis, drop analysis, principal component analysis, cluster analysis, and multiple regression analysis were used to classify their cold tolerance types, establish cold tolerance prediction models, and screen evaluation parameters.

【Result】

The variation of each parameters at normal conditions were minor fluctuations ranging from 3.12% to 18.89%. The seedling emergence rate was above 85.00%, which had high viability and could be used for subsequent cold tolerance analysis. The variability of each parameter within the accessions increased under chilling stress, ranging from 7.14%-108.33%, and the most variable parameter were root length under CC condition and germination index under DVC condition. Principal component analysis converted the 14 parameters under chilling stress and 100-grain weight measured into six mutually independent composite indicators, representing 74.98% of the total data information. The comprehensive cold tolerance evaluation value (D) was calculated by the affiliation function method and then clustering analysis was performed. 200 cotton varieties (lines) were divided into five categories according to their cold tolerance, with 2 of the group Ⅰ being strongly cold tolerant, 42 of the group Ⅱ being cold tolerant, 69 of the group Ⅲ being medium cold tolerant, 83 of the group Ⅳ being more sensitive, and 4 of the group Ⅴ being sensitive, of which Xinluzhong 16 was the most cold-tolerant material. A multiple regression analysis was applied to establish a prediction model for cold tolerance of cotton at seedling emergence as Y=-4.10+0.58X4+0.40X14+0.32X1+0.22X5 (R2=0.92), and four parameters for cold resistance evaluation were confirmed, namely total length of seedling, emergence rate, and dry weight under CC stress, germination rate under DVC stress. The cold-tolerant varieties (lines) had higher seedling emergence rates of early sowing experiment in the field, which were basically consistent with the results of the indoor results.

【Conclusion】

It is feasible to use CC and DVC stress combined with multivariate statistical analysis to evaluate the cold tolerance of cotton at seeding stage, and total length of seedling, emergence rate, and dry weight under CC stress, germination rate under DVC stress can be used as evaluation parameters.

【Objective】

To establish an evaluation system for low phosphorus tolerance in cotton varieties (lines), screen low phosphorus tolerant cotton germplasm and evaluate different types of phosphorus efficiency, and lay the foundation for studying the physiological mechanisms of low phosphorus tolerance in cotton and mining low phosphorus tolerance genes.

【Method】

Using 140 cotton cultivars (lines) from different cotton regions at home and abroad, 21 traits such as biomass, root-related indexes and phosphorus efficiency-related indexes were measured under low (10 μmol·L-1 KH2PO4) and normal (500 μmol·L-1 KH2PO4) phosphorus treatments in a hydroponic experiment. The index of low phosphorus stress tolerance was calculated for each index. Using the integrated affiliation function method, principal component analysis, regression analysis and cluster analysis were conducted to classify the low phosphorus tolerance of each cotton variety and to comprehensively evaluate the low phosphorus tolerance and phosphorus efficiency type of each cotton variety.

【Result】

Compared with the normal phosphorus treatment, the mean values of total phosphorus accumulation, total phosphorus content, aboveground dry weight and total dry matter weight of the tested cotton varieties decreased more under the low phosphorus treatment, while the mean values of root average diameter, specific root area, root tips number and phosphorus use efficiency increased. Under low phosphorus treatment, the coefficients of variation of each index ranged from 6.04% to 47.79%,the coefficients of variation of root indexes such as specific root tips density, root tips number, specific root length and root average diameter were higher than those of normal phosphorus treatment, and the coefficients of variation were 47.49%, 42.13%, 40.19% and 19.16%, respectively; the principal component analysis of the 21 indexes of low phosphorus stress tolerance showed that the cumulative variance contribution of the six principal components reached 77.21%, and the comprehensive low phosphorus tolerance value (D) was calculated using the affiliation function method. The D-value regression equation was established by multiple regression analysis to determine the six low phosphorus tolerance indices and perform systematic clustering to classify different cotton varieties (lines) into three categories: low phosphorus tolerant, intermediate and low phosphorus sensitive.

【Conclusion】

Total dry matter weight, phosphorus use efficiency, root fresh weight, total root length, root surface area and total phosphorus accumulation were identified as indicators for the evaluation of low phosphorus tolerance in cotton.

Early ABA-stimulated maintenance of Cl homeostasis by mepiquat chloride priming confers salt tolerance in cotton seeds

Mepiquat chloride (MC) priming alleviates the effects of salt stress during seed germination in cotton (Gossypium hirsutum L.), but the mechanisms underlying its effects are unknown. We found that MC priming increases salt tolerance, as evidenced by marked increases in seed vigor and germination rates, and alleviated salt toxicity by reducing Cl accumulation in germinating seeds. Consistently, electrophysiological experiments revealed that the seeds with MC priming displayed superior Cl exclusion ability in the root apex. These beneficial effects of MC priming were abolished by the abscisic acid (ABA)-synthesis blocker fluridone under salt stress. MC priming induced an early response to acclimatization and stress, as indicated by rapidly increasing ABA content during initial exposure to salt stress. Transcriptome analyses revealed that MC priming induced an array of differentially expressed genes (DEGs) in germinating seeds. The most noticeable changes in germinating seeds were MC priming-induced increases in the expression of DEGs encoding components of ABA biosynthesis, ABA catabolism, and ABA signaling pathways under salt stress. MC priming also increased the expression of some DEGs encoding Cl ion transporters (e.g. CCC, SLAC1/SLAH1/SLAH3, CLC, and ALMT9) in germinating seeds. These results indicate that MC priming-induced ABA contributes to Cl homeostasis in tissues and acts as a positive regulator of salt tolerance via regulation of Cl transporters (particularly CCC and SLAC1/SLAH1/SLAH3). Taken together, these findings shed light on the molecular mechanism underlying MC-mediated tolerance to salt stress during seed germination.
