• Lactobacillus rhamnosus MP108 relieved ulcerative colitis in mice.
• L. rhamnosus MP108 improved the intestinal barrier by increasing the number of goblet cells, MUC2 production and tight junction protein expression as well as inhibiting epithelial cell apoptosis.
• L. rhamnosus MP108 inhibited colonic inflammation by regulating the balance of pro-inflammatory and anti-inflammatory cytokines.
• L. rhamnosus MP108 elevated the relative abundance of short-chain fatty acids producers, causing an increase in acetate and butyrate levels.
