Highlights
• The treatment of L-theanine and geniposide (TG) effectively alleviated depression-like behaviors and hippocampal neuron damage induced by CUMS in mice.
• RNA-seq sequencing revealed that the Transthyretin (Ttr) gene was upregulated in the hippocampus of CUMS-induced depression-like mouse models, while treatment with TG reduced its expression.
• TG treatment improved inflammation, apoptosis-related gene and protein expression, and neurotransmitter levels in the hippocampus of CUMS-induced depression-like mouse models.
• TG treatment affected changes in the gut microbiota, indicating its potential as an effective therapy for depression-like behaviors in mice.
• Depression can upregulate Ttr expression in the hippocampus, which may be involved in inducing depression-like behavior. TG treatment can effectively alleviate depression-like behavior through multiple pathways.
