Highlights
• We identified a counter-trend expansion of a fibroblast subpopulation (Fib_5: ADAM12+ COMP+ POSTNhi) exhibiting a markedly fibrogenic phenotype in hypertrophic scars (HTSs), consistent with conserved pro-fibrotic cellular states observed in other fibrotic skin disorders.
• Our findings demonstrated that YY1 downregulation exerts pro-fibrotic effects in HTS fibroblasts, while its overexpression attenuates fibrogenic activation, highlighting YY1 as a key transcriptional regulator of fibroblast fibrotic programs.
• We revealed significant changes in fibroblast–pericyte signalling in HTS, which was associated with multiple fibrosis-related pathways through cell–cell communication analyses.
