abstract = {Plant tissue culture is an asexual propagation technique based on the principle that plant cells are totipotent. This technique is widely used in plant propagation, breeding and genetic engineering. Although many colleges and universities offer plant tissue culture experiments in undergraduate experimental courses, the plant materials used are usually limited to larger plants such as tobacco, which are over 60 cm tall, and the experimental period is long. The root segments of the model plant Arabidopsis thaliana are easy to induce callus and easy to observe. Therefore, this paper investigates the feasibility of opening A. thaliana tissue culture experiments in undergraduate experimental teaching. As demonstrated by several undergraduate and graduate students in our laboratory, the experiment is easy to operate and observe, requires a short time to obtain results and has good reproducibility. And by setting different ratios of auxin and cytokinin, it can help students better understand the important role of plant hormones in plant tissue culture.}
