abstract = {【Objective】Eucalyptus urophylla×E. grandis, recognized as a significant timber species, possesses extensive applications and considerable economic value. In this study, cloning, bioinformatics analysis, subcellular localization and tissue-specific expression profiling were employed to investigate the nuclear transcription factor EuNF-YB1. This research provides a theoretical basis for understanding its basic characteristics and its regulatory role in the growth and development of Eucalyptus urophylla×E.grandis.【Method】EuNF-YB1 was obtained from DH3229 clones of Eucalyptus urophylla×E.grandis at 6 months of age by PCR. The clones were analyzed by bioinformatics, subcellular localization and tissue-specific expression.【Result】EuNF-YB1 gene coding region was 534 bp, encoding 178 amino acids with a molecular weight of 19.10 kD and pI 6.44, which belongs to the unstable hydrophilic proteins. Multiple sequence alignment analysis showed that EuNF-YB1 also contained conserved DNA-binding domains, NF-YA interaction domains and NF-YC interaction domains, indicating that EuNF-YB1 was also a typical NF-YB transcription factor. Evolutionary relationship analysis showed that EuNF-YB1 was closely related to AtNF-YB1 and CmNF-YB8 protein. The results of subcellular localization showed that the protein was localized in the nucleus and cytoplasm. Tissue-specific expression analysis showed that EuNF-YB1 gene was expressed in all tissues, with the highest expression in flowers.【Conclusion】The cloning and expression analysis of the EuNF-YB1 gene in Eucalyptus urophylla×E. grandis indicated that this gene may play a significant role in the growth and development of Eucalyptus, particularly in the regulation of flowering.}
